ABSTRACT:
The integration of chemometric tools with advanced spectroscopic techniques has revolutionized pharmaceutical analysis by enabling high-throughput, non-destructive, and environmentally sustainable assessments of drug quality, authenticity, and process efficiency. This review critically examines the evolution, application, and future trajectory of chemometric methodologies, including PCA, PLSR, ANN, and SVM, in deciphering spectral data within the pharmaceutical domain. Emphasis is placed on the role of big data, preprocessing strategies, model validation metrics (e.g., RMSE, R²), and data mining techniques for maximizing interpretability and prediction accuracy. Spectroscopic methods, including UV-Vis, IR, Raman, NIR, fluorescence, and NMR, are explored in tandem with chemometrics for their ability to handle complex matrices and overlapping signals in formulations, APIs, and herbal products. The review further highlights practical case studies and real-time Process Analytical Technology (PAT) implementations that demonstrate chemometric applications in drug stability testing, counterfeit detection, and phytopharmaceutical authentication. Limitations such as multivariate model complexity, data reproducibility, and software dependency are critically analyzed, while emerging trends like deep learning integration, regulatory alignment with QbD frameworks, and AI-enhanced model development are discussed as transformative directions for the field. By consolidating current literature and analytical paradigms, this review serves as a robust reference for researchers aiming to leverage chemometrics in pharmaceutical quality control and innovation.
